1. Famílies de receptors i mecanismes d’activació de la transcripció
Els receptors nuclears són factors de transcripció activats per lligand. En general són
l’últim component d’una cascada. Com que el que fan és regular l’expressió gènica,
tenen efecte a llarg termini. Els receptors nuclears només es troben als animals.
Els receptors nuclears s’envolten de proteïnes coactivadores, que potencien la
transcripció del gen regulat, i de proteïnes corepressores, que la inhibeixen.
Els receptors nuclears no produeixen cascada de senyalització, però s’ha vist que els
lligands que reconeixen receptors nuclears poden interaccionar amb altres cascades,
donant lloc a les anomenades accions no genòmiques dels receptors nuclears. Un
exemple d’aquest fet és la progesterona i el seu receptor.
This schematic shows the interaction of the
classical progesterone receptor (PR) with Src-
homology-3 (SH3) domains and the initiation of
the extracellular signal-regulated kinase
(ERK)/mitogen-activated protein kinase (MAPK)
cascade. The amino-terminal proline-rich
domain of PR interacts with the SH3 domain of
Src. This activates the Ras–Raf–ERK/MAPK
pathway, which then influences the activity of
transcription factors (TFs) in the nucleus. The
more conventional (genomic) action of the PR is
depicted on the right of the figure; PR functions
as a dimer in the nucleus to mediate
transactivation and subsequent transcriptional
