De synthese van cholesterol is een complex proces 18 molecule acetylCoA worden verbruikt om
uiteindelijk een cholesterolmolecule met 27C-atomen te maken
De basisstappen:
STAP 1: De synthese van de mevalonaat (5C-atomen) uit acetylCoA via het intermediar
hydroxymethylglutarylCoA (HMG-CoA)
STAP 2: De synthese van isopreen-bouwsteen isopentenylpyrofosfaat (IPPP) uit mevalonaat
STAP 3: Polymerisatie van 5 van deze geactiveerde bouwstenen tot squaleen
STAP 4: Ringsluiting van squaleen tot cholesterol
FUNCTIE
Cholesterol = belangrijk bestanddeel van het lipidenmembraan stabilisator
Een cel kan deze cholesterol uit de omgeving halen, of zelf synthetiseren vanuit AcetylCoA
Voor sommige cellen is er nog extra cholesterol nodig, omdat de cellen het intensief nodig hebben
voor de productie van bv. steroïdhormonen of vitamine D
REGELING
Snelheidsbeperkende stap = reductie van HMGCoA tot mevalonaat gekatalyseerd door HMG
CoA reductase
Enzym wordt allosterisch sterk geremd door de vrije cholesterolconcentratie in de cel en
inhibeert daardoor zijn eigen synthese
Statines courant gebruikte cholesterolverlagende geneesmiddelen zijn synthetische remmers van
dit enzym
Wanneer de novo synthese van cholesterol wordt geremd lichaam cholesterolreserves
aanspreken om die op te gebruiken
Er wordt dus meer cholesterol van de periferie naar bv de lever gehaald onder de vorm van
HDL-cholesterol + het totale cholesterolgehalte daalt
