Phoebe - Introduction – We are presenting the paper “MicroRNA-132 enhances transition from
inflammation to proliferation during wound healing”. We have both researched the different four
stages of wound healing: Haemostasis, inflammation, proliferation and maturation, and the
specificity of micro RNA 132’s role within the process.
Mark – Li, D et al have specifically researched the role of epidermal keratinocytes and miRNA-132
within the process of wound healing. Each stage of wound healing overlaps, however Li and
colleagues suggest that miRNA-132 aids in the transition from inflammation to proliferation.
Phoebe - To study micro RNA expression during human skin wound healing, they created an in vivo
study of 14 healthy volunteers. They made surgical wounds and collected the tissue around the
wounds at different time points after injury that were selected to represent the sequential phases of
skin wound healing: haemostasis (0 hour), inflammation (24 hours), proliferation (1 week), and
remodelling (1 month). As the graph shows primary miRNA-132 and miRNA-132 show a spike 1 week
into the healing process indicating the role within the inflammatory process
Mark – The main objectives of the paper encompassed the aim to broaden the knowledge of the
role of micro RNA’s within wound healing and the global effects on the transcription and regulation.
There is potential for micro RNA’s to be used therapeutically in the future if we can understand
more.
Phoebe – The results of the investigation show TGF-β induces miR-132 expression in peripheral
wound keratinocytes. miR-132 demonstrates two functions through the regulation of HB-EGF and
other targets. It can decrease NF-kB activity and restrict inflammation, but also increase the activity
of STAT3 and ERK, thus promoting cell growth.
Mark – Overall the paper was a remarkably interesting and informative paper, we decided from a
reader’s perspective with a fair amount of understanding around the topic, the paper seemed well
structured but overcrowded. All the information was relevant to the topic however, the research
was extremely broad in nature. A better paper would have had a clearer and more concise topic in
mind rather than several actions of the micro RNA.
Phoebe – We concluded that the research carried out by Li, D and colleagues was hugely beneficial
to understanding the role of micro RNA’s in wound healing and has provided a good insight into how
miRNA-132 can have global effect on the human body when healing. We think the next step for
micro RNA research will be to look at therapeutic routes to treat chronic wounds such a diabetic
ulcers now we understand their biochemical role.
