, Abstract
Het eerste ontdekte fluorescerende eiwit is Green Fluorescent Protein (GFP) en werd in 1962 ontdekt
door Dr. Shimomura bij de kwal Aequorea Victoria. GFP is in staat groen licht uit te zenden dat zich in
het spectrum van zichtbaar licht bevindt. Het fotoprotein aequorin bevindt zich in de kwal en kan
door binding met calcium blauw licht uitzenden, dit kunnen De Green Fluorescent Proteins
vervolgens omzetten in groen licht. Het bestaan van fluorescente eiwitten heeft vele nieuwe
inzichten mogelijk gemaakt in de geneeskunde en biochemie. In dit onderzoek wordt onderzocht
voor welk fluorescerende eiwit het plasmide codeert. Er zijn vier verschillende plasmiden die elk
coderen voor een ander fluorescent eiwit. Deze kunnen als volgt coderen voor YFP (Yellow
Fluorescent Protein), RFP/mCherry (Red Fluorescent Protein), GFP (Green Fluorescent Protein) of CFP
(Cyaan Fluorescent Protein). Om erachter te komen welk fluorescent eiwit het betreft, is er gebruik
gemaakt van DNA- en eiwitanalyse. Eerst werd de bacteriestam Escherichia Coli vermeerderd en
geïsoleerd. Voor de DNA-analyse is gebruikt gemaakt gelelektroforese en voor de eiwitanalyse
SDS-page en fluorescentie- en absorptiespectrometrie. Uit de verkregen resultaten kan worden
geconcludeerd dat het plasmide codeert voor mCherry (Red Fluorescent Protein).
