In dit werk is de HPLC-DAD gebruikt om een nieuwe methode te ontwikkelen voor de bepaling van
morfine, codeïne, 6AM, cocaïne, BEG, cocaethyleen, methadon en zijn metaboliet EDDP in plasma
, door gebruik te maken van Bond Elut Certify-cartridges om de medicijnen uit de biologische vloeistof
te extraheren. [1]

structuurformule in een bepaalde matrix een keuze maken uit een
voorbewerkingsmethoden zodanig dat deze analiet geanalyseerd kan worden
met een chromatografische techniek.
1.4.1 Geef de analiet met naam

1.4.2 Geef de chemische en physische eigenschappen van het molecuul met verwijzing literatuur
Cocaïne (methyl (1R,2R,3S,5S)-3- (benzoyloxy)-8-methyl-8-azabicyclo[3.2.1] octane-2-carboxylate) is
een vaste stof met de molecuulformule C17H21NO4. De structuurformule van cocaïne staat in figuur 1
gegeven. Het moleculaire gewicht is 303,35 g/mol. De logP is 2.3 wat laat zien dat deze stof apolair is
en de pKa is 8,61. Cocaïne heeft een kookpunt van 187 °C en verder heeft cocaïne een oplosbaarheid
in water (22°C) van 1800 mg/l. [2]

1.1.1 Beschrijf de matrix en karakteristieken van de matrix
In het artikel bevindt de cocaïne zich in bloedplasma. Geneesmiddelen worden vaak in plasma
bepaald, omdat dit type monster zorgt voor een goede correlatie tussen doses die zijn toegediend en
gedetecteerde niveaus. Een breed scala aan analytische methoden voor het kwantificeren van
drugsmisbruik in plasma met behulp van gaschromatografie of hogedrukvloeistofchromatografie is de
afgelopen jaren gemeld.
Plasma is de extracellulaire matrix die de bloedcellen bevat. Het maakt 55 procent van het totale
bloedvolume uit. Het bestaat voor 95% uit water en voor 8% uit eiwitten. Het eiwit zoals
serumalbuminen, globulinen en fibrinogeen.[3] Daarnaast bevat het ook glucose, elektrolyten en
stollingsfactoren. PH van plasma ligt tussen 7,35 en 7,45.[4]

Alle monsters werden onderworpen aan vaste-fase-extractie door middel van Bond Elut Certify-
patronen volgens het volgende protocol:
1: kolom conditionering met 2 ml methanol en 2 ml fosfaatbuffer bij pH 6; langzame toevoeging van 1
ml plasma. Voor de bond elut certify-cartridges is de conditionering met 2 mL methanol en 2 mL buffer
iets standaard. De fosfaatbuffer wordt gebruikt omdat deze buffer zich ook bevindt in de mobiele fase
gradient. De methanol dient om de cartridge te reinigen en activeren. Bij het werken met
serum/plasmamonsters is buffer van 1-2 ml een typisch volume voor het verwijderen van resterende
MeOH in de conditioneringsstap. Dit voorkomt eiwitprecipitatie wanneer het monster door de cartridge
gaat
