• Paden in de ontwikkeling van huidkanker:
- Huidkanker heeft twee ‘’pathways’’ wat samen leidt tot kanker:
o P53 mutatie
o Verminderd immuunsysteem (door de zon)
- Uitleg:
o Je DNA brokkelt af waardoor er mutaties komen in de keratinocyten
o P53 is een tumor suppressor cel die ontregeld raakt waardoor er kanker kan
ontstaan
o Naast UVB is ook elke expositie tot UVA verantwoordelijk voor huidkanker; UVA
radiatie produceert mutaties in het DNA, dit vormt mutaties in het P53 tumor
suppressor gen
o Mutaties in het P53 gen resulteren in een non-functioneel proteïne, wat
gemuteerde keratinocyten kan herstellen
o Normaal gesproken zou een beschadigde cel geprogrammeerd zijn tot
geprogrammeerde celdood (apoptose)
o Dit leidt dus tot een ongecontroleerde celgroei en proliferatie van de losgeslagen
cellen (maligniteit)
o Ook zorgt de zon ervoor dat het lokale immuunsysteem van de huid minder
wordt
• Meest voorkomende lokalisaties van kanker in 2015:
- Als het incl. basaalcelcarcinoom was geweest, dan zou huidkanker staan op nummer 1
• Het voorkomen van verschillende soorten kanker:
- BCC:
o 70%
o >62 jaar
o 0% metastaseert
- PCC:
o 20%
o >75 jaar
o 2% metastaseert
- Melanoom:
o +/- 10%
o >52 jaar
o 20% metastaseert
• Incidentie (aantal nieuwe gevallen per jaar):
- BCC: 35.000 per jaar
- PCC: 9.500 per jaar
- Melanoom: 5500 per jaar
