Metaboliet profiling: Nuclear Magnetic Resonance (NMR), Liquid chromatography
(LC), gas chromatography (GC) en mass spectometry (MS).
LC/MS: individuele chemische componenten in aparte pieken, niet-gasvormende
stoffen worden ook gevonden.
Pre-processing -> peak tables met pieken per specifiek metaboliet en zijn
isotopen.
Piek hoogte en oppervlak: informatie over de relatieve concentratie
LC-MS data figuur: x-as: tijd. Y-as: intensiteit. Z-as: m/z waarden.
Total Ion Current (TIC): som van alle intensiteiten van alle m/z waarden (alle
massa’s laat hij zien, overall chromatography duidelijk)
Extracted Ion Chromatograms (EIC of XIC): som van intensiteiten van één of
meerdere m/z waarden (schone chromatogrammen, duidelijk te zien pieken,
potentiele isomeren duidelijk)
Identificatie van pieken:
Je verkrijgt dan piek positie, hoogte, oppervlakte en integratie grenzen -> peak
table
Aanpak: 1. Two directions: identificatie van pieken op de m/z as en de retentie
tijd 2. XIC/EIC slices: m/z as in kleine slices -> voor elke m/z slice een nieuwe
chromatogram (=EIC) 3. Model fitting: 2D gaussian curve op de data leggen.
We bespreken alleen optie 2.
EIC per bin -> peak detection op elke EIC.
