El ciclo de Krebs es una ruta anfibólica: participa en procesos catabólicos y anabólicos. El
ciclo proporciona α-cetoglutarato y oxalacetato para la síntesis de glutamato y aspartato
respectivamente, entre otras moléculas fundamentales para la célula.
La reacción de oxidación - decarboxilación del piruvato es el nexo entre la glucólisis y el ciclo
de Krebs. Esta reacción irreversible es catalizada por un complejo enzimático (piruvato
, deshidrogenasa) localizado en la matriz mitocondrial de eucariotas, y en el citosol de
procariotas.
El piruvato pierde el grupo carboxilo como CO2, y los dos carbonos restantes unidos a la
CoA conforman la acetil-CoA. En la reacción se reduce un NAD a NADH.H que a su vez cede
los H a los otros transportadores de cadena respiratoria, con la consecuente formación de 3
ATP.
En cuatro reacciones del ciclo ocurren oxidación de intermediarios y reducción de coenzimas
de cadena respiratoria: tres NAD y un FAD. Esas moléculas reducidas que integran la
cadena respiratoria se reoxidan, y parte de la energía liberada se usa para fosforilar el ADP a
ATP. En el ciclo propiamente dicho se produce una fosforilación a nivel de sustrato que
produce un GTP, que equivale energéticamente a un ATP.
El ciclo se puede resumir en la siguiente ecuación:
Acetil-CoA + 3NAD + FAD + GDP + Pi CoA-SH + 3 NADH.H + FADH 2 + GTP + 2 CO2
