Instructions: For this lab activity, you will examine a section of an onion root tip under a
compound light microscope to determine the approximate amount of time spent in each stage of
a cell’s life cycle. At the end of your lab activity, you will complete chi-square testing using some
of your data. Submit your lab report to your instructor when completed.
Title: Finding the Time Spent in Different Stages in the Cell Cycle
Objective(s): This lab serves the purpose to do the following: to see and record data in
order to infer things about a cell’s life cycle (as to which stages are the most prominent), to
use the chi squared test in order to test the quality of the data, overall the lab serves to have
us infer things based on the data that we collect.
Hypothesis: If the stage has been recorded the most times then it must be the most
prominent.
Write a hypothesis, using an if/then statement, to predict the stage in which a cell spends most of
its time.
Procedure:
Access the virtual lab and complete both examinations of onion root tip cells. The materials are
listed for you. However, you are responsible for providing a brief summary of the steps you
followed during the experimental procedure.

1. Prepare slides for examination:

c. Stain them in order to make the chromosomes more visible
d. Cover each specimen with a cover slip
2. Then classify each cell based on what phase it is in
The phases to be classified are as follows:
- Interphase: The DNA duplicates to prepare for mitosis. The chromatin appears as
a tangled mess
- Prophase: The chromatin in the nucleus condense to become visible under the
microscope as chromosomes. The nuclear membrane dissolves and the spindle
fibers form
- Metaphase: The spindle fibers align the chromosomes along the middle of the
cell’s nucleus
- Anaphase: The paired chromosomes separate and move to opposite sides of the
cell
- Telophase: New membranes each set of chromosomes while the chromosomes
spread out into chromatin
- Cytokinesis: The cell physically divides into two separate daughter cells.
3. Count the cells found in each phase
4. Use these numbers to predict how much time a dividing cell spends in each phase
