, Q1. Make a temporary, stained preparation of T.S of sunflower stem. (4 marks)
 Slide preparation – 3 marks (NOT TO WRITE ON THE ANSWER SHEET)
o Take 8 to 10 thin sections of the sunflower stem given and put them in a watch glass containing water.
o Select and transfer one thinnest section in another watch glass containing dilute safranin and let it stain for
1 -2 mins.
o Take a clean slide and put a drop of glycerine in the centre of the slide.
o With the help of a brush transfer well stained section on the slide. Place a coverslip avoiding any air bubbles.
o Blot excess of glycerine from the sides of the coverslip. Observe the slide under 10X objective of microscope
 Draw labelled diagram of the examiner’s choice out of the following on the answer sheet: (1mark)

6. Is sunflower stem dicot or monocot? Why? Dicot (because vascular bundles are open, in a ring and not
scattered as in monocot)


Q 1. Mitosis in onion root tip (4 marks)
OR
 Squash preparation – 2 marks (NOT TO WRITE ON THE ANSWER SHEET)
o Take a root tip in watch glass and wash it with water two times. Drain off the water
o Add 1 N HCl to the watch glass and warm it over the spirit lamp.
o Remove from the flame and let it cool for some time.
o Remove the HCl and wash the root tip two times with water.
o Take the root tip on a clean slide.
o Cut and keep the milky white portion of the root tip on the centre of the
slide, discard the remaining part.
o Put a drop of Acetocarmine stain.
o Let the root tip remain in acetocarmine for 20-25 minutes.
o After 20-25 mins put coverslip on it and squash it.
o Observe first under 10x (low power) and then under 45x (high power).
 Identification of any stage of mitosis under the microscope - 1 m
