2. Protein Composition and Structure
Eiwitten zijn de meest veelzijdige macromoleculen in levende systemen en vervullen cruciale
functies in essentiële biologische processen. Ze functioneren als katalysator, transporteren andere
moleculen, zorgen voor mechanische steun en het immuunsysteem, zorgen voor beweging, geven
impulsen door en leiden groei en differentiatie.

- Eiwitten zijn lineaire polymeren die opgebouwd zijn uit monomeren (aminozuren). De
volgorde heet de primaire structuur. Hieruit wordt een 3D-structuur gevormd. De functie van
eiwitten hangt af van de primaire structuur.
- Eiwitten bevatten een brede selectie van functionele groepen.
- Eiwitten kunnen inwerken op een ander eiwit of op andere biologische macromoleculen
zodat een complexe verzameling gemaakt kan worden.
- Sommige eiwitten zijn stijf, andere flexibel. Stijve eiwitten zorgen voor structuur in
cytoskeleten of bindweefsel.

2.1 Proteins Are Built from a Repertoire of 20 Amino Acids
Aminozuren zijn de bouwstenen van eiwitten. Een α-aminozuur heeft een centraal, chiraal
(asymmetrisch) koolstofatoom, met daaraan een aminogroep, een carbonzuur, een waterstofatoom
en een restgroep.

Van eiwitten bestaat door het chirale koolstofatoom een L en een D isotoop. Alleen de L variant
bestaat als eiwit, doordat deze beter oplosbaar is, en vermoedelijk dominanter daardoor.

Aminozuren in een neutrale omgeving, komen meestal voor als dipolaire ionen (zwitterion). De
aminogroep is hierbij geprotoneerd (-NH3+) en de carboxylgroep is gedeprotoneerd (-COO-). In een
zure omgeving is de aminogroep geprotoneerd en de carboxylgroep is neutraal (-COOH). Wanneer de
pH stijgt, geeft de carboxylgroep als eerste een proton weg, wanneer de pKa rond 2 ligt. De
aminogroep staat een proton af, wanneer de pH rond 9 ligt.

Eiwitten hebben twintig verschillende zijgroepen. Ze worden ingedeeld in vier groepen:
1. Hydrofobe aminozuren met niet-polaire restgroep. Glycine is het enige hydrofobe
aminozuur dat niet chiraal is. De driedimensionale structuur van water-oplosbare eiwitten
wordt gestabiliseerd door de neiging van hydrofobe groepen om naar elkaar toe te gaan;
hydrofoob effect.
2. Polaire aminozuren met neutrale restgroep, maar de lading is niet gelijk verdeeld. Drie van
deze aminozuren hebben een hydroxyl als zijgroep (-OH). Deze -OH-groep zorgt ervoor dat
het aminozuur nog meer hydrofiel en reactief wordt. Er bestaan ook aminozuren met -SH in
plaats van -OH. Deze zijn nog reactiever, omdat ze zwavelbindingen kunnen vormen.
3. Positief geladen aminozuur met een positieve restgroep. Dit zijn aminozuren die positief
geladen zijn bij een neutrale pH.
4. Negatief geladen aminozuur met een negatieve restgroep. Deze aminozuren hebben
meestal een zijgroep dat een proton mist, zijn meestal zuur, en daarom dus negatief geladen.

Zeven van de aminozuren hebben gelijk ioniseerbare zijgroepen. Deze kunnen zowel protonen
afstaan als opnemen, reacties vergemakkelijken en ionbindingen vormen.
