Summary (Samenvatting):
The goal of this experiment is to determine the concentration quinine in tonic with different
methods. The used methods are: the standard addition method, and the external standard method.
The spectrofluorometer is used for the standard addition method and the HPLC-UV-FLU is used for
the external standard method. The used monsters are: royal club tonic and royal club tonic with a
unknown concentration of NaBr. Also the quenching constant of broom is determined with a Stern-
Volmer plot. The results of this experiment are: the quenching constant of broom is: 460,8. The
concentration of quinine in tonic is: 44,4 ± 1,802. The concentration quinine in tonic with a unknown
concentration of NaBr is: 43,4 ± 4,546.
Doel:
Het doel van dit onderzoek is het bepalen van de concentratie kinine in tonic met verschillende
methodes. De verschillende methodes zijn: de standaard additie methode met als analyse apparaat
de spectrofluorometer en een externe standaard methode met HPLC scheiding en met UV en
Fluorescentie detectie. Als monster is royal club tonic gebruikt en royal club tonic met een
onbekende concentratie NaBr. Verder is ook de quenching constante bepaald met behulp van het
Stern-Volmer plot. En is er een 95% betrouwbaarheidsinterval opgesteld voor de concentratie kinine.
Hypothese:
De concentratie kinine in tonic bedraagt: 60 mg/l [1]
De excitatiegolflengte van kinine is: 310 nm* [2]
De emissiegolflengte van kinine is: 450 nm* [2]
*quinine sulfate in 0,5 M H2SO4.
De quenching constanten van chloor bedraagt: 209,95 [3]

Aan het begin van dit onderzoek zijn de excitatie en emissie golflengte van kinine bepaald met
behulp van de FP6300 spectrofluorometer en de SHIMADZU UV-1280 en UV-1800. De gevonden
excitatiegolflengte bedraagt: 247 nm. De gevonden emissiegolflengte bedraagt: 452 nm.
