Elke spier bevat volgende delen
1. de gebruikelijke celorganellen (bv.: mitochondriën)
2. sarcolemma = celmembraan ⇒ geleiding v/d AP
3. contractiel apparaat = dikke en dunne myofilamenten ⇒ contractie
Dit apparaat is opgebouwd uit sarcomeren ⇒ functionele eenheid waarbij myofilamenten
elkaar organiseren. De interacties tussen de dunne en dikke filamenten zorgt voor
spiercontractie.
1. Dikke filamenten
- opgebouwd uit myosine: staart en kop
, - tijdens contractie gaan de kopjes hechten aan de actine op de dunne
filamenten
2. Dunne filamenten
- actine: actieve plaats (eiwitten) waar
myosine zal op reageren
- tropomyosine: in rust zal dit de actine
verbergen
- troponine (eiwit): houdt tropomyosine
op zijn plaats + hoge affiniteit om calcium te binden
Noch dikke, noch dunne filamenten overspannen heel de lengte v/e sarcomeer. De dikke
filamenten liggen in het midden v/e sarcomeer. De dunne filamenten zijn aan beide uiteinden
v/e sarcomeer vastgehecht aan eiwitten die Z-lijn vormen. De rangschikking van dikke en
dunne filamenten geven het gestreept uiterlijk.
- Z-lijn: eiwitten die uiteinden v/d dunne filamenten vasthechten
- M-lijn: eiwitten die dikke filamenten verbinden
- H-zone: zone waarin geen dunne filamenten liggen
- A-band: zone waarin dikke filamenten liggen
- I-band: zone waarin geen dikke filamenten liggen
Titine en nebuline zijn gigantische hulpeiwitten. Titin overspant de afstand van een Z-schijf
tot de naburige M line (titin helpt dus bij de relaxatie van de spieren). Nebuline, liggend langs
de dunne filamenten, hecht zich aan een Z-schijf maar strekt zich niet uit tot de M-lijn. Het
helpt de actine uit te lijnen.
4. sarcotubulair systeem ⇒ excitatie-contractie koppeling
- lengterichting: sarcoplasmatisch reticulum
- gespecialiseerde vorm ER
- verwijding op het einde tot cisternen
(voorraad voor calcium)
- dwarsrichting: T-tubuli
