Geschiedenis
Rond 1900 was syfilis (spirocheen) een groot probleem. Arseen gebruikt als GM = vroege
chemotherapie. Arseen is toxisch dus het was sterven aan het GM of aan de ziekte. Magische kogel
genoemd ➔ dood de kiem maar niet de dierlijke/humane cel. Dachten ze, was niet waar.
Fleming met de ontdekking van penicilline = introductie van AB. Onderzoek op stafylococcen. Plaat
besmet met schimmel ➔ Penicillium Notatum. Bacterie rond de schimmel kunnen niet groeien ➔
eerste AB afkomstig van deze schimmel. Het duurt nog een periode voor het op de markt komt. Vroeger
nog geen goed ontwikkelde technologie. Zorgden in de oorlog dat soldaten hun wonden overleefden.
Eenmaal op de markt werd het vrij verkocht ➔ veel patiënten namen een onderdosering ➔ veel
bacterie resistentie tegen penicilline.

Fase van de PG synthese in S. Aureus
Eerste fase: cytoplasma
Start: NAG met UDP aan. Enkel schrijven:
Mur-enzyme ➔ NAG omzetten tot NAM.
UDP niet kunnen tekenen. Hier
stafylococcus aureus = gram pos ➔ 5 AZ
aan vast tekenen. Eind resultaat: NAM,
UDP en 5 AZ. Gebeurt in het cytoplasma.
Tweede fase: membraan
Doorheen het membraan brengen.
Onderaan membraan laten binden ➔
bactoprenol (C55) ➔ fosfaat hangt eraan
vast, zit in het membraan (lipofiel). Anker
waarbij de fosfaatgroep aan de binnenkant
zit. NAM gaat aan het anker hangen ➔
UMP = monofosfaat groep, gaat weg. Er
moet een NAG aankomen + op de Lysine
moeten nog de 5 glycines komen =
zijketen. Monomeer nu aan de buitenkant
van het membraan. Enkel weten gebeurd
door een enzyme (Flippase).
Derde fase: extracellulair
Monomeren met elkaar verbinden: vormen
een lange suikerketen. NAM/NAG verbonden
met elkaar. Dimeer komt los van het
membraan. Dit is de eerste stap.
Transglycosylase ➔ alle monomeren aan
elkaar hangen. Transpeptidase = tweede stap
➔ 3-4 peptide binding maken. Reacties niet
kunnen tekenen.
