Een posttraumatische-stressstoornis (PTSS) wordt in de DSM-5-TR omschreven als een

stoornis die kan ontstaan na het ervaren van, geconfronteerd worden met, of het getuige zijn van

traumatische gebeurtenissen, waarbij de persoon feitelijk of dreigend overlijden, ernstig letsel of

seksueel geweld ervaart. PTSS kan zowel door eenmalige, als door langdurige en herhaalde

blootstelling aan traumatische gebeurtenissen ontstaan. De symptomen kenmerken zich als

herbelevingen, vermijding, negatieve verandering in cognitie en stemming, en verhoogde

prikkelbaarheid. Om aan de diagnostische criteria te voldoen, moeten de symptomen minimaal één

maand aanhouden en aanzienlijk lijden en disfunctioneren veroorzaken (Taylor-Desir, 2022). Hoewel

niet elke traumatische gebeurtenis tot PTSS leidt, wordt de levensprevalentie tussen 8% en 12%

geschat in de algemene bevolking (Brooks & Greenberg, 2024) en 7% in Nederland (VZinfo, 2016).

PTSS is een complexe aandoening die vaak in verband staat met psychische comorbiditeit, zoals

suïcide en alcohol- en middelengebruik (Mitchell et al., 2023). De huidige behandelingen voor PTSS

bestaan uit diverse psychotherapieën, waarbij voornamelijk blootstelling aan de traumatische

gebeurtenis centraal staat, zoals Eye Movement Desensitization and Reprocessing (EMDR) en

Exposure therapie. Evenals farmacologische behandeling, voornamelijk selectieve

serotonineheropnameremmers (SSRI) (Illingworth et al., 2020). Echter blijft een aanzienlijk deel (35-

47%) van de patiënten na behandeling aanhoudende symptomen houden. Ook wordt PTSS in

verband gebracht met grote sociaaleconomische kosten en ziekteverzuim (Mitchell et al., 2023).

Vanwege de grote individuele en maatschappelijke impact, zijn effectievere therapeutische

behandelingen nodig (Illingworth et al., 2020).

Steeds meer onderzoek wordt gedaan naar ondersteunende behandeling met 3,4

methyleendioxymethamfetamine (MDMA) bij volwassenen met PTSS. Onderzoek bij kinderen en

jongeren met MDMA en PTSS is nog nauwelijks gedaan, en verreist een ander
