The differentially expressed mRNA, also known as differential gene expression (DEG)
is the analysis and interpretation of differences in abundance of gene transcripts within a
transcriptome. This is different from the gene expression in which the genetic information is
stored in the gene is converted into functional gene product preferably in the form of protein
but non coding RNA in some instances.
Analysis of differentially expressed mRNA:
Mainly there are three technologies widely been used in the analysis of differentially
expressed mRNA viz. microarray, High throughput Sequencing of cDNA (RNA Sequencing) and
Differential Expressions using Customised Amplification Libraries (DECAL).
High throughput Sequencing of cDNA (RNA Sequencing):
Compared to other technologies such as microarrays, direct sequencing of RNA
provides a more comprehensive understanding of transcriptome complexity and the ability
to detect a dynamic range of expression levels. Generally, high throughput sequencing of
cDNA includes direct sequencing of complementary DNAs (cDNAs) by high-throughput next
generation sequencing (NGS) technologies. This is followed by the Mapping of the sequencing
reads to the reference genome or gene sets for gene expression analysis and polymorphism
detection and Library preparation.
Differential Expressions using Customised Amplification Libraries (DECAL).
Differential expression using customized amplification libraries (DECAL), is a technique
which permits global comparisons of gene expression under varied growth conditions without
a specific requirement for DNA arrays. The key feature of DECAL technology is the ability to
amplify by PCR a complex mixture of expressed genes in a reproducible and representative
manner without the confounding effects of rRNA or any other highly expressed gene product.
