“Pre-Lab Questions”
1. Which DNA nitrogenous bases pair with each other? Which bases are purines, and which are
pyrimidines?
DNA nitrogenous base pairs with each other are:
Adenine and Guanine are purines
Cytosine and Thymine are pyrimidines

2. How is DNA information used to make proteins? What are the steps of this process?
DNA is used to make protein through the process of transcription and translation. When
enzymes from DNA molecules are transcribed into mRNA with RNA polymerases. The
next mRNA reaches the ribosome, then tRNA and translates into amino acids that block
proteins.

3. Give an example of a scenario in which you would perform PCR vs a scenario in which you would
use recombinant DNA technology.
You would perform PCR when determining an illness and would be used to get DNA
from a blood sample. When using recombinant DNA technology it can be used for
human growth hormones such as insulin.

4. What occurs during each of the three steps involved in the PCR cycle? How has the use of PCR
changed biotechnology?
The three steps (parts) to the PCR cycle are: Denaturing, Annealing, and Extending.
During denaturing the mixture is heated to separate the DNA strands. Next is the
annealing phase where it is cooled enough to facilitate hydrogen bonds to form new
separated DNA strands. Lastly the temperature is raised again to allow DNA polymerase
to use the primer-DNA hybrids to synthesize two new DNA molecules. PCR has
changed biotechnology, in that now many viral and bacterial infections can now be
identified with PCR screenings.

5. How could you take a protein with a known sequence of amino acids and use it to create an
artificial gene?
A protein with a known sequence of amino acids can be used to create an artificial gene
by using recombinant DNA technology with bioinformatic tools.

“EXPERIMENT 1: DNA Extraction
Post-Lab Questions
1. What is the purpose of the following reagents in the experiment?
