Introductie tot stamcellen
functionele definitie
● stamcellen beschikken over de capaciteit tot zelfhernieuwing = delen en opnieuw stamcel worden
● stamcellen kunnen gedifferentieerde dochtercellen voortbrengen bij deling
→ stamcel op zich is niet functioneel
● stamcellen hebben bij celdeling keuze tot het voortbrengen van één stamcel en één gedifferentieerde
dochtercel = asynchrone deling
Stamcellen evalueren (criteria)
zelfvernieuwing
● extensieve in vitro proliferatie (> 160 populatieverdubbelingen)
● geen oncogene transformatie
● op dit moment enkel aangetoond voor embryonale en neurale stamcellen (zie grafiek)
○ moeilijk te bereiken in lichaam
clonaliteit
● één enkele stamcel moet in staat zijn meerdere stamcellen voort te brengen
● ≠ polyclonale stamcelpopulatie rechtstreeks geïsoleerd uit weefsel

Nut van stamcellen

voorbeeld: hematopoietische stamcellen (HSC)
● zelfhernieuwing van HSC geeft nieuwe HSC om toekomstige hematopoiese te ondersteunen
● differentiatie van HSC resulteert in het ontstaan van snel prolifererende hematopoietische
progenitorcellen (HPC), die vervolgens alle bloedceltypes kunnen produceren
Adulte stamcellen
afhankelijk van het ontwikkelingsstadium verschillende types stamcellen
● primordial germ cells
○ ontwikkelen onafhankelijk van kiembladen
○ vormen later gameten
● placental stem cells
○ bron van stamcellen → invriezen als donor aan andere kindje
○ niet gebruiken bij ziekte op jonge leeftijd (aangeboren)
○ te weinig voor adult
● adulte stamcellen = stamcellen voorkomend na de geboorte
○ komen voor in bijna alle weefsels
○ dragen bij tot natuurlijke weefselvernieuwing en/of herstel na beschadiging
