slide
2. with the inoculating loop, aseptically add bacterial culture to the drop of
water
3. mix the bacteria with the water and spread it out
4. allow suspension to air dry
5. heat-fix the suspension (smear) by quickly passing the slide through a flame
three times.
Gram Stain Procedure Right Ans - 1. Prepare bacterial smear
2. Add Crystal violet for 1 minute
3. Rinse slide w/water then add mordant (iodine) for 1 minute
(something that helps fix dye on or in a cell but does not itself contribute
color)
4. rinse slide w/water
5. decolorize w/ Gram Alcohol by dripping down the slide until color flushes
out, rinse with water
6. Counterstain w/ Safranin for 2-3 minutes then rinse w/ water and blot dry
Acid Fast Stain Right Ans - 1. apply primary stain- carbolfuchsin
4- 5 min (rinse w/ h2o)
2. decolorize with acid-alcohol (dripping it) until color runs out then rinse w/
water
-non acid-fast bacteria decolorize,
acid-fast remain pink
3. apply counterstain- methylene blue (1 minute)
rinse w/ h20
-acid-fast are pink, non acid-fast are blue
4. blot dry
* Use Mycobacterium as positive control
Endospore Stain Right Ans - 1. Prepare bacterial smear * Bacillus +
2. Use Malachite green to flood slide and add paper towel- ensure its saturated
3. Use heat as a mordant. Flame the slide until steam appears (4-5 passes)
