In preparation for the lab day, please read the lab in its entirety and fill out the following sections. Thank
you. This ELN is due Wednesday Oct 2nd no later than 11 am in the appropriate assignment folder on
A2L. Please submit one pdf file of this completed template. Please note: it is your responsibility to
ensure that your submitted files are not corrupt and can be properly opened or your submission will be
incomplete. Additionally, you must submit in the appropriate assignment folder on A2L otherwise your
submission will be incomplete.
Purpose
In the space provided below:
1. Purpose - Briefly describe the purpose of this lab. What is your overall goal for this lab day (very
concise, cannot exceed 500 words)?
To perform directional cloning to ensure that the amplified folA gene is properly oriented with
respect to the T7 promoter in the pET26b. An agarose gel will be synthesized during the lab in order
visualize the DNA bands. By the end of this lab, I will gain experience in working with restriction
enzymes, preparing an agarose gel, and in loading DNA samples. As a result, I will be able to gain an
understanding in interpreting DNA gels.
