For this practical you will need:
● A light microscope;
● An onion;
● Iodine (or any stain);
● A slide;
● A coverslip;
● A ruler;
M e t h o d
1. Cut your onion and peel a very thin layer of it.
2. Place your onion sheet on a microscope slide and make
sure that it is sitting straight on it, without forming any
bubbles.
3. Put 3 drops of iodine or any other stain on your onion
layer and cover it with a coverslip.
4. Use the coarse focusing knob on your microscope to
distance the stage from the objective lens.
5. Clip the slide onto the stage and adjust the coarse focus
so that the objective lens are almost touching the slide.
Look at the microscope from the side, at this step, or you
might risk damaging the slide.
