Samenvatting op basis van mijn notities, de slides en het boek. Achteraan zijn er een aantal oefenvragen terug te vinden die al dan niet op Sofia staan. Voor de richtingen Biomedische Wetenschappen en Tandheelkunde, deze behandelen dezelfde leerstof. :))

Verloop
Stap 1 (voorbereidende fase)
= hexokinasereactie
● In kort: glucose wordt op plaats 6 gefosforyleerd ter vorming van glucose-6-P
○ Investering van ATP
● Door hexokinase
○ = isoenzym, familie van hexokinases, hierin ook glucokinase

, ○ Komt in meeste weefsels voor
■ Niet in lever en bèta-cellen daar glucokinase
○ Lage Km, dus hoge affiniteit ⇔ glucokinase hoge Km
○ Lage Vm ⇔ glucokinase hoge Vm
■ ‘Wat is nu het nut van deze verschillende parameters?’
● Hersenen moeten altijd goede voorraad aan glucose hebben,
wanneer schaars in lichaam gaat dit naar orgaan met grootste
opp = lever ⇒ niet goed voor hersenen, daarom verschillende
hexokinases in lever
● ‘Waarom’
○ Gradiënt behouden, anders [glucose] in cel te hoog waardoor geen transport
meer naar binnen
○ Non-ionisch glucose wordt omgezet in een anion wat niet door de membraan
terug kan naar buiten + geen glucosetransporters voor glucose-6-fosfaat ⇒ zit
gevangen in cel
○ Aanzet tot meer reactieve molecule glucose
○ Fosforylatie regelt hoeveel glucose er verder gemetaboliseerd mag worden
Stap 2 (voorbereidende fase)
= fosfoglucose/-hexose isomerase reactie
● In kort: van 6- naar 5-ring, van gesloten naar open vorm ter vorming van
fructose-6-fosfaat
● Isomerase verplaatst aldehydefunctie naar C2 in open vorm, wanneer deze gesloten
wordt krijgt met een 5-ring
○ Van aldose naar ketose suiker
● Reversibele stap, lage ΔG, wordt niet geregeld
