Werking:
Gezuiverd eiwit PITC aan toevoegen, bestaat uit 2 delen: aromatische ring (helpt achteraf de
detectie: absorbeert licht) en de reactieve chemische groep (bindt aan de amino terminus van een
eiwit door een nucleofiele aanval)
Dan pH verandering shuffling van elektronen uiteindelijk gaat het laatste aminozuur
afgesplitst worden van het eiwit en gaat hangen aan de aromatische groep die te meten is
HPLC (high performance liquid chromatography) doen elk AZ heeft een andere retentietijd: obv
dit kan je zien welk aminozuur afgesplitst is
Dit dan 50x doen
Zeer omslachtige sequentiebepaling; 40-60 AZ per peptide nu geautomatiseerd; eiwitsequenties
opgeslagen in databanken bv Swiss-Prot.
Sequentie dikwijls afgeleid van DNA-databanken. Meer en meer sequentiebepaling via massa
spectrometrie
1.2. Eiwit sequentiebepaling via massa spectrometrie
2 grote strekkingen: MALDI-TOF vs MS/MS
Beginnen met een gezuiverd eiwit (door kolomchromatografie of via 2D-gel dus scheiding volgens
lading en volgens massa)
Stap 1: Eiwit knippen met protease Bij voorkeur trypsine (alle peptides gaan 2 dezelfde ladingen
hebben):
o Endopeptidase trypsine knipt aan C-terminale zijde van Arg & Lys (hebben positieve lading)
alle peptides gaan dan eindigen met een positieve lading
o levert peptiden van ≤ 20 AZ
o peptiden hebben een C-terminaal basisch residu
o na ionisatie: 2 pos ladingen: NH2 terminaal en C-terminaal
eenvoudiger spectrum als alle peptiden een dubbele lading hebben (spectrogram:
rangschikking volgens massa)
