Serotonine (5-HT) komt voor in de hersenen (onder andere in de hypofyse als precursor van
melatonine); in de entero-chromaffine cellen in het gastro-intestinale systeem (90% bij de mens);
zenuwcellen myenterische plexus; orthosympatische zenuwuiteinden; nociceptieve zenuwuiteinden;
en in trombocyten.
Productie
De eerste stap is de snelheidsbepalende stap. 5-
hydroxytryptamine, ofwel serotonine kan worden opgenomen
in bloedplaatjes. Hier kan het een ringmethylatie ondergaan
tot 5-methoxytryptamine. We kunnen 5-HIAA meten, want dit
is een maat voor de serotonine afbraak bij een normaal dieet.
Receptoren
Er zijn verschillende serotonerge receptoren (5-HT 1 tot en met
5-HT7). Iedere receptor heeft zijn eigen weefseldistributie en
farmacologie. De 5-HT3 receptor bestaat uit vijf subeenheden
rond de centrale kationen. Ook 5-HT 4 is selectief voor kationen
en dus betrokken bij de excitatorische synaptische transmissie.
Serotonerge synaps
Wanneer de actiepotentiaal de terminus bereikt van het
presynaptische neuron, zullen de vesikels fuseren met de
membraan. Serotonine zal vrijkomen in de synapsspleet en zal
binden met de postsynaptische receptoren. Nu wordt er een
actiepotentiaal gegenereerd in het postsynaptische neuron.
Wanneer serotonine in het postsynaptische neuron komt,
wordt het via MAO gerecycleerd. De serotonine in de
synapsspleet kan naar de bloedbaan gaan; maar wordt ook
weer verwijderd uit de synapsspleet (door uptake-1 of de presynaptische 5HT 1 receptor) om
desensibilisatie te voorkomen voor nieuwe pulsen van serotonine. Serotonine zelf zal ook een
receptor activeren die de vrijzetting van serotonine onderdrukt. Deze neurotransmitter doet dus aan
zelfinhibitie.

Erg variabel in functie van species en fysiologische toestand, wat veralgemeningen
bemoeilijkt
Enorm aantal receptoren, waarvan voor een aantal zelfs de fysiologische functie ongekend is

Buspirone wordt gebruikt bij angst
Sumatriptan wordt gebruikt bij migraine
Cisapride wordt gebruikt bij reflux
