Typing
A. Blood Typing
1.Understand the four blood types in the ABO blood group. Understand which antigens
are on the surface of RBCs and which antibodies are in the plasma for the ABO blood
group system.
2.Understand how the blood typing procedure works, and be able to determine blood
type by observing blood typing results (clumping or no clumping). Recognize the
,antisera used in blood typing –anti-A, anti-B, and anti-D (Rh).
3. Understand the transfusion compatibilities for the ABO blood group.

1.Understand that a blood smear is done using Wright’s stain.
- Wright's stain is a histologic stain that facilitates the differentiation of blood cell
types. It is classically a mixture of eosin (red) and methylene blue dyes.
2.Be able to identify and name erythrocytes and platelets and be able to distinguish
between and name the three types of granular leukocytes and the two types of
agranular leukocytes.
- Erythrocytes are red blood cells
- Platelets are tiny blood cells without a nucleus that helps with clotting.

A. Hemoglobin Content using a Hemoglobinometer
1.Recognize the hemoglobinometer and understand how it is used.
- Turn ON the hemoglobinometer.
Wait until you see the number code in the display (for example 950) that corresponds to
the
test card number (found on the bottle).
Wait until the flashing “test card” symbol is visible to insert the white test card into the
hemoglobinometer. You will feel it “click” into place.
Wait until you see the “blood drop” symbol to add a small drop sterilized sheep’s blood in
the
center of the test card. Be careful not to overload the well.
Wait until the reading is stable. This may take a minute. Read and record the value in the
machine’s display
Remove the white test card by lifting up slightly and pulling it out
2.MEMORIZE the normal hemoglobin value ranges for males and females, and be able
to determine whether given readings are normal or abnormal.
- Normal hemoglobin for males are 13.0-18 g/dL & females are 11.5-16 g/dL
