, First Identification Test: Gram Stain
The image of the gram stain shows an organism that is spherical in shape which indicates
that the organism is cocci or coccus. The image shows an organism that is a shade of red. Since
the organism is not a shade of purple that means the organism is gram-negative.
Based on the decision tree the next test I want to run would be the capsule strain.
Second Identification Test: Capsule Strain
In the capsule strain test, we will prepare a microscope slide with thin smears of the
bacteria, let it air dry, apply 1% crystal violet allowing it stay for 2 minutes, gently remove the
crystal violet with 20% copper sulfate, do not use water to rinse, and then visualize with
microscope (Western Governors University, 2020)
In the image of this organism during the capsule strain, it shows the organism with a dark
background. This organism is not showing any space around the cell. The lack of space around
the cells tells me that the cell is negative for being encapsulated, which means that this organism
does not form a capsule around organism. If the organism had space around the cell, it would
indicate the organism having a capsule, but this organism does not indicate this.
Based on the decision tree, the next test I want to perform the glucose fermentation test.
Third Identification Test: Glucose Fermentation Test
The phenol red broth tubes will be prepared, add the sample being test, add the Durham
tube (inverted tube), aseptically transfer the pure bacteria culture to a sterile tube of phenol red
