Alle andere reagentia die worden gebruikt in dit hoofdstuk zijn niet gevaarlijk of worden in dergelijk
lage concentraties of volumes gebruikt en worden als dusdanig niet als gevaarlijk beschouwd.
Samenvatting: Er wordt altijd een labojas en veiligheidsbril gedragen in het laboratorium. Bij het
hanteren van Ampicilline (100 µg mL-1 ) worden handschoenen gedragen om huidreacties te
voorkomen. Wegens een zeer felle allergie aan ampicilline werd ook een isolatieschort en
mondmasker gedragen. Bij het gebruik van en E. coli XL1 Blue worden handschoenen gedragen
omdat het over mutanten gaan. Vermijd de lozing van en E. coli XL1 Blue in het milieu.
Procedure
Voor de procedure wordt verwezen naar de handleiding op pagina 57 en 58.
Bij het oppikken van de kolonie werd ook het tipje van de micropipet mee in 5mL LB gedaan. Voor
stap 2 hebben we niet gewacht tot een OD van 0,8. We gingen verder bij een OD van 0,587. Deze
waarde zou in orde moeten zijn want de cellen zouden dan in exponentiële fase moeten zijn.
Berekeningen en waarnemingen
Waarnemingen: Na de groei, die overnacht gebeurde waren de bacteriecellen troebel. De optische
densiteit bij 600nm is 0,12.
