MICROSCOPE
Microscope is a laboratory equipment that allows scientists to view minute objects and specimen, by proving a
magnified look, to visualize its composition that cannot be seen with the naked eye. They are made up of lenses of
different magnification, which have their own powers, this will allow the eyepiece lens to focus on an object clearly, to
its focal strength. (Mokobi, 2021) [2]

EQUIPMENT
 Optical Microscope: An optical microscope is used to observe, the
different samples such as hair and glass
 Light source: allows the sample to be seen much more clearly.
 Microscopic slide (cavity slides): These were used as they have a
dense spot, so the samples are still and do not move.
 Forceps: are used to prevent cross-contamination of samples.
 Sample.

METHODOLOGY:
Observe the slide under the microscope and identify the composition and
properties of each sample and compare.

HAIR
A piece of hair can be solid evidence in forensic examinations and is known as
‘good’ evidence this because they are sturdy and do not degrade which means
they can survive for period of times. This is because they contain a protein
called keratin which makes it resistant to wear and tear. Hairs also carry a lot of
biological information. They are also easy to examine and cost effective.

HAIR STRUCTURE
The hair grow from root and projects through skin. The roots are embedded in
the skin and have a follicle (blood supply). The follicle cells produce new cells
which move away from the skin this cuts off blood supply and nourishment and
produce keratin protein which hardens the structure. Cells begin to change and
become flatten, line up and squeeze together into packed layers and then lose their nuclei and fills with protein giving
cells strength and resistance.

I. CENTRE PORTION – Medulla (appears dark under microscope) is the central part of hair. It is a canal like structure
of cells and runs through centre of cortex. Some hair may not have a medulla or may have fragmented medulla
(e.g., human head hair). Animal hair has thicker medulla which takes up more than two thirds the width of the hair

II. CORTEX – Contains microscopic features and can help determine species. Bulk zone of the hair underneath the
cuticle and is made up from spindle shaped cortical cells aligned in a helical arrangement with air spaces present
(known as cortical fusi). The cortex is randomly embedded with oval shaped melanin pigment granules. When new
hair is being produced, melanocyte cells in the hair follicle produce melanin. Hair becomes white/grey with age
because melanocyte cells stop working so pigment does not form.

III. CUTICLE – layered scales covering the shaft that can help determine whether hair is animal or human. Has a clear
and protective outside covering and gives hair its resistance to chemicals. It also allows the hairs to retain its
structural features over a long period of time. Formed by separate overlapping scales that point inwards towards
the tip. Tip may not have scales – fragments chip off the ends causing ends to split.
