Sustaining Proliferative Signaling
Kankercellen kunnen ongecontroleerd groeien doordat ze het vermogen hebben om proliferatieve signalen te
manipuleren. Normale weefsels reguleren zorgvuldig groeibevorderende signalen om celgroei en -deling in balans te
houden. Deze signalen worden voornamelijk overgebracht door groeifactoren die binden aan celoppervlaktereceptoren
en intracellulaire signalen activeren. Het begrip van deze processen in normale weefsels blijft echter beperkt. In
tegenstelling hiermee is de mitogene signalering in kankercellen beter begrepen. Kankercellen kunnen proliferatieve
signalen op verschillende manieren behouden, zoals het produceren van hun eigen groeifactoren, stimuleren van
omringende normale cellen om groeifactoren te leveren, verhoging van receptoreiwitten of constitutieve activering van
signaalwegen. Deze manipulaties stellen kankercellen in staat om onafhankelijk van externe groeifactoren te groeien en
kunnen verschillende downstream-signaalwegen activeren, wat bijdraagt aan hun ongecontroleerde groei.

Somatic mutations activate additional downstream pathways
DNA-sequencinganalyses van kankercelgenomen hebben ontdekt dat somatische mutaties in bepaalde menselijke
tumoren leiden tot constitutieve activering van signaalcircuits, meestal geïnitieerd door geactiveerde
groeifactorreceptoren. De voordelen van het activeren van upstream (receptor) versus downstream (transducer)
signalering voor tumorcellen zijn nog niet volledig begrepen, evenals de functionele impact van de communicatie tussen
