Define chronic asthma
Chronic inflammatory condition of the airways, hyper responsiveness
Define acute asthma
Progressive worsening of symptoms. With exacerbations resulting in marked
reduction in baseline lung function
Describe arachidonic acid metabolism to leukotrienes
Arachidonic acid > (5 lipoxygenase) > Leukotriene A4
What do they do?
Leukotrienes bind to CysLT1/2 receptors on SM
Causes contraction
Increases vascular permeability
Increases leukocyte activation
How could aspirin increase the risk of asthma?
Blocks COX so causes all the arachidonic acid to be used by 5 lipoxygenase to
be turned into leukotrienes to cause bronchoconstriction
Define COPD
Chronic obstructive pulmonary disease
Smoking impairs lung defences leading to recurrent bronchitis and pneumonia
The inflammatory reaction increases tissue destruction
How is it treated?
Inhalers - salbutamol
MoA Salbutamol
Agonist on B2 adrenoceptor on smooth muscle cells
Gs → increased adenylate cyclase → increased cAMP → increased PKA
