Detailed summary notes for week 2 topic 2 (pharmacokinetics and pharmacodynamics) of the 7PAYFADD; PSYCHOLOGY & NEUROSCIENCE OF ADDICTION module at King's college London .
W2 T2; Pharmacokinetics and pharmacodynamics
Psychopharmacology – how drugs change mood, cognition and behaviour
Our focus is psychoactive drugs that affect CNS; therapeutic use (anti-epileptic) and
recreational (addiction)
Current lack of understanding on MOA; synapse level understanding but not whole
brain
Neuronal network and receptor interactions are complex so add to difficulty
Pharmacokinetics
What our body does to drugs
These parameters affect drug use:
o A – administration/absorption
o D – distribution
o M – metabolism
o E – extraction
Administration
Method of taking drug:
o Oral (tablet)
o Injection; intramuscularly (IM), intravenously (IV) and subcutaneous (SC)
o Inhalation
o Application to skin
o Mucous membranes; intranasal (snorting), sublingual (under tongue)
Route of admin depends on chemical characteristics of drug, personal preference,
how long drug remains in body and dosage (concentration profile)
E.g. below shows different peaks, concentration and duration
Absorption is how quickly drug can pass cellular barrier into blood stream
IV is fastest as it is direct; in all the rest, need to cross at least one cellular membrane
