Growth of yeast in liquid culture

Yeast can be grown in either liquid medium or on the surface of a solid agar plate.
Like bacteria, yeast cells reproduce asexually.This is a new cell that eventually breaks away
from the parent cell.

Aim
To see how temperature affects growth of yeast

Equipment

- 3 Water baths at (30℃, 40℃, and 50℃)
- Yeast in liquid culture
- 4 250ml Flasks
- Stirring rod
- pipette
- Stiker label with marker
- Colorimeter and 6 colour filter
- Cubettes for the colorimeter

Method

1. Before the experiment was begun, the water baths were switched on and prepared to
the needed temperature
2. The yeast was divided into four 250 flasks, each labelled with different temperatures,
room temperature (20℃), 30℃, 40℃, and 50℃
3. Then the colorimeter was set and prepared, 6 colour filter were inserted, a cuvette
was taken and was filled till the marked line with water and placed inside the
colorimeter
4. The absorption was set to 100, with the dial
5. Then the flask containing the yeast solution, was stirred with the stirring rod, then
was poured into a cuvette with the use of a pipette
6. The absorption was then recorded for each different temperature, ensuring that the
water was inserted and set to 100, before measuring each temperature
7. This was all then repeated, to increase the accuracy of the data
8. After every hour the absorption was measured and recorded
9. Then the next morning it was also measured and recorded
