Setting up/ using a light microscope
Handling the light microscope
1. With two hands, I carried the microscope to the center of the desk.
2. I plugged the microscope into the closest plug socket, ensuring the cord was out of the
way as much as possible.
3. I next santised the work desk with alcohol to ensure there was no chance any
bacteria/fingerprints present could touch the slide and interfere with the specimen
analysed.
4. Furthermore, when handling any slides with the specimen on it, I always grasped the
slide from its edges to ensure no fingerprints were touching the specimen.
Using the light microscope
1. I first screw/clicked the lowest power objective lense into place.
2. Then I lowered the stage considerably low using the coarse adjustment knob.
3. Next, holding the slide appropriately, I placed the slide center on the stage (ensuring the
specimen was placed above the hole in the stage)
4. I then moved the stage clips over the end of each slide to hold the slide into place,
looking through the eyepiece to ensure the specimen is in the centre of the stage.
5. Next I raised the stage up using the coarse adjustment knob and turned the light on the
microscope.
6. Looking through the eyepiece, I adjusted the distance of the stage from the objective
lense using the coarse adjustment knob (ensuring the lenses are not touched).
7. After the specimen was in focus using the coarse adjustment I focused onto the
specimen more by using the fine adjustment knob.
8. I then increased the magnification of the specimen by ensuring the microscope was still
focused at its lowest objective lense (generally 4X) before increasing the microscope to
a higher magnification.
9. I did this by rotating the next largest magnification lens into place and refocusing the
specimen using the fine adjustment knob.
10. When finished using the microscope I turned the light off and unplugged the microscope.
11. I then lowered the stage using the cIoarse adjustment knob and turned the nosepiece so
the lowest objective lens is clicked into place.
12. Next I removed the stage clips from the slide and gently removed the slide and stored it
appropriately.
13. Then I wrapped the cord around the base of the microscope and with two hands I carried
the microscope back to where it is stored.
