Practical One


Growth Media This is the medium used for microbial growth.
Usually agar is used for this.

Defined Growth Media:
Known concentrations of known ingredients.

Complex Growth Media:
Undefine growth media, rich in nutrients of which the chemical
composition is exactly known.

Aseptic Technique ● Disinfect and sanitize all laboratory surfaces.
● Petri dish lids must not be open for extended periods
of time.
● When opening the petri dish lid, keep it close to the
base to prevent entry of microbes.
● Do not breath near/onto sterile instruments.

Inoculation Apparatus 1. Inoculation loop
2. Inoculation needle
3. Sterile toothpick

Transferring Bacteria from 1. Light the bunsen burner and flame inoculation loop for
Liquid Culture to Solid sterilisation. (Do not set it down once sterile).
Medium 2. Open test tube (containing liquid bacteria) and run
mouth of test tube through flame.
This ensures no microbes present on the neck of the
tube enter and contaminate the sample.
3. Place the inoculation loop inside the liquid culture and
remove it.
4. Flame the tube mouth again before closing it.
5. Run the inoculation loop over the solid medium to
transfer the liquid culture. Turn the petri dish to ensure
every quadrant is covered.
6. Close the petri dish lid.
7. Flame the inoculation loop.

Transferring Bacteria from 1. Light the bunsen burner and flame inoculation loop for
Liquid Culture to Liquid sterilisation.
Medium 2. Open test tube (containing liquid bacteria) and run
mouth of test tube through flame.
3. Place the inoculation loop inside the liquid culture and
remove it.
4. Flame the tube opening again before closing.

and flame mouth.
6. Place the inoculation loop into the liquid medium.
7. Flame the neck of the test tube containing liquid
medium and close.
8. Flame the inoculation loop.

Transferring Bacteria from 1. Sterilize inoculation needle on bunsen burner flame
Solid Medium in Petri Dish to and let it cool.
Solid Medium 2. Rub inoculation needle over a single colony in the
petri dish.
3. Open test tube containing solid medium and transfer
the colony by making three deep stabs.
4. Close test tube.
5. Flame needle to sterilize.

Transferring Bacteria from 1. Sterilize inoculation loop on bunsen burner flame and
Petri Dish to Liquid Medium let it cool.
2. Rub inoculation loop over a single colony in the petri
dish.
3. Open test tube and flame the mouth.
4. Place inoculation loop into the liquid medium.
5. Flame the mouth and close the test tube.
6. Flame inoculation loop.
