Practical Notes

Part 1-Intended learning outcomes (ILOS)
• Explain how to isolate bacteria and yeast from environmental samples
• Explain the underlying principles of culture purification
• Define the term colony and colony morphology (in terms of microbiology)
• Describe a microbial colony in terms of its morphology
• Evaluate both the macro and micro-morphology of bacteria and yeast

Colony morphology
→ In microbiology, colony morphology refers to the visual appearance of bacterial or fungal colonies on an
agar plate.
→ Examining colony morphology is the first step in the identification of an unknown microbe.
→ The systematic assessment of the colonies' appearance, focusing on aspects like form, size, elevation,
margin, surface texture, opacity and colour provides clues to the identity of the organism, allowing
microbiologists to select appropriate tests to provide a definitive identification.

• Colony~ a cluster of identical cells grown on the surface of solid medium originated of a single mother
cell.
→ Different types of bacteria can form different types of colonies
→ Colonies belong to one species may differ in their shape, size, colour and texture from others.
→ Surface can be described as: smooth/glistening/rough/wrinkled/dull

▪ FORM- the basic shape of the colony
▪ SIZE- the diameter of the colony
▪ ELEVATION- describes the side view of the colony
▪ MARGIN- the edge of the colony
▪ SURFACE- appearance of the surface of the colony
▪ OPACITY- describes the transparency
▪ COLOUR- describes the colour (pigmentation)

TJW Notes

, It is important to remember that the colony morphology of a single species can CHANGE depending on the
type of medium used to culture it.
